Scientists have uncovered a new target for treating skin inflammation and related diseases. The G protein-coupled estrogen receptor 1 (GPER1) plays a key role in slowing down the excessive growth of skin cells, known as keratinocytes. Recent research suggests this receptor could lead to more effective and safer therapies for conditions like psoriasis and atopic dermatitis.
The findings, published in the Journal of Investigative Dermatology in 2023, highlight GPER1's potential in reducing inflammation and improving skin health.
The research team, led by Pérez-Escudero, conducted experiments on human keratinocytes and fibroblasts in the lab. They found that activating GPER1 with compounds like G1 significantly slowed cell proliferation, particularly during the G1 phase of the cell cycle. This effect was linked to the receptor's ability to interfere with the MAPK/ERK pathway, lowering the production of pro-inflammatory mediators such as TNF-α and IL-6.
In animal studies, mice with psoriasis-like skin inflammation were treated topically with a GPER1 agonist for seven days. The results showed a clear reduction in disease severity, including less inflammation and epidermal thickening. Unlike traditional estrogen receptors, GPER1 provides rapid cellular responses, offering a distinct way to modulate skin inflammation without the broader side effects often seen with hormone-based treatments.
The study also emphasised GPER1's connection to estrogen signalling, though its mechanisms differ from those of classical nuclear receptors. This distinction could allow for more selective therapies, targeting inflammation while minimising unwanted effects on other tissues.
The discovery of GPER1's role in skin cell regulation opens doors for new treatments. Pharmacologists may now develop GPER1-targeted drugs, either as topical creams or systemic medications, to manage chronic skin conditions. Additionally, measuring GPER1 levels in patients could help tailor therapies, improving outcomes in personalised medicine.
